Abstract

In this study, the antioxidative effects of carboxymethyl-chitooligosaccharide (CM-COS) on the generation of direct free radical scavenging activity was examined using an electron spin resonance (ESR) spectrometer and assess the cytotoxicity of CM-COS. Antioxidative activities of deacetylated COS at 70% (CM-COS-70) and 80% (CM-COS-80) were measured, respectively. In the ESR analysis, the CM-COS-80 was found to have the highest antioxidant activity when compared to the other CM-COS. The radical scavenging activities (effective concentration at 50%, EC50 (mg/mL)) of the CM-COS-80 were 3.29, 6.25, 2.65 and 1.50 mg/mL for DPPH, hydroxyl, alkyl and superoxide, respectively. Moreover, CM-COS did not show any cytotoxic effect against the MRC-5 cell line. In addition, the CM-COS-70 and -80 significantly inhibited oxidative damage to purified genomic DNA. Therefore, CM-COS derivatives may be used as a source of antioxidants, potentially as a food supplement, or by the pharmaceutical industry.
